Leaders Arsenal, currently six points ahead and having played one additional match, will visit the Etihad Stadium on Sunday at 16:30 BST.
During an appearance on The Wayne Rooney Show, he commented that Manchester City "understand how to secure the title" and "have a manager familiar with the process".
"I believe City has the advantage in that regard, primarily due to their manager and the talent they possess. They are likely to maintain a more composed demeanor than the Arsenal squad," Rooney stated.
"They might feel anxious because when losing several games in a row, thoughts shift to 'where will the next goal or win come from?' That kind of negative thinking significantly affects overall performance."
On Sunday, Manchester City secured a 3-0 victory over Chelsea, making the most of Arsenal's unexpected 2-1 loss to Bournemouth on Saturday.
Rooney mentioned that whichever team triumphs in Sunday’s "title decider" in Manchester will become the frontrunner for the league championship.
Furthermore, he expressed that he believes a draw could be considered a "solid point" for Arsenal, who may need to take a more aggressive approach.
He remarked that Mikel Arteta’s team should "be difficult to penetrate, ensure there are no gaps in their defense, and look for counter-attack opportunities".
He added: "Regardless of whether the fans appreciate that style of play, they must do whatever it takes to prevent City from winning. They need to demonstrate resilience to keep City from scoring."
'Would be a disaster' if Spurs are relegated
Tottenham Hotspur finds themselves at the bottom of the league after their loss at Sunderland combined with West Ham's victory over Wolves.
Rooney described Spurs' display in the northeast as "a bit lackluster," which hindered their ability to gain momentum under new manager Roberto de Zerbi.
"There are times when teams benefit from a boost after a managerial change, but that usually happens when the new head coach instills a proactive, aggressive style," he explained. "I felt they were somewhat caught in between."
The former England forward mentioned that De Zerbi must "quickly find solutions" and emphasized that relegation to the Championship would be catastrophic for the club.
"It would be a disaster for them if they do get relegated," Rooney stated. "Many of those players might be contemplating that going down could lead to opportunities with other clubs.
"If they fail to return to the Premier League in the first attempt, that would be an absolute nightmare for them."
Tottenham has not secured a win in the Premier League in 2026, and their next match will be at home against Brighton on Saturday at 17:30.
Rooney emphasized that they must devise a strategy to achieve victory to lift their morale.
"If they succeed in this, they enhance their chances. The longer their winless streak lasts, the more challenging it becomes for them."
